Research

Proteins have revolutionized the treatment of many chronic and life-threatening diseases. On the other hand, proteins are sensitive compared to traditional drugs. This research is part of the EU project RealHOPE where the effect of real-life handling is studied. The focus in my research is subvisible particles from aggregation triggered by different stressors during transport and handling. Subvisible particles can trigger immune reactions leading to anaphylaxis in severe cases but also loss of efficacy due antibodies against the drug. During the first part the transport inside a hospital to the wards of the cancer drug trastuzumab was studied. Here the use of pneumatic tube transport was found to release particles from the materials in the IV bags used and without protective measures the stress during transport triggered protein aggregation. One important finding was that when the air headspace was removed the particle formation was reduced. In another part, the effect of stress that can occur during handling for example shear stress in syringes, and interactions with materials used in medical devices, including silicone oil lubricants, will be investigated.
