Description

The Microbial Flow Cytometry Labs (established in 2014) is located at the Division of Applied Microbiology, which is a part of the Center for Applied Life Sciences (CALS) at the Department of Chemistry (KILU), LTH. It is well integrated in current activities within education, research and industry co-operations. The current user group consists of LU researchers, other Swedish universities, and national and international companies.

Equipment and resources

The Microbial flow cytometry labs have 1 cell sorter and 2 analytical bench flow cytometers (see specifications below). The placement of the infrastructure enables access to other available Resources at the Division of Applied Microbiology and Kemicentrum, making it possible to perform controlled microbiological experiments and sensitive post-sort analyses.

Beckman Coulter Moflo XDP cell sorter. Lasers: violet (405nm), blue (488nm) and yellow (561nm) Output: FSC, SSC, and 10 filters for measurement of fluorescense emission. Analysis mode: Up to 100 000 events/s, and resolution <0.2μm to 25μm Sorting mode: Up to 70 000 events/s, max 4-way sort simultaneously into sample tubes, microtitrer plates, petri dishes. Sample port: Tubes with different sizes (0.5-50 ml)

BD Accuri C6 Csampler. Lasers: blue (488 nm), red (640 nm). Output: FSC, SSC and 4 filters. Sample port: sip can be applied for any size (Eppendorf tubes to bioreactors). Csampler : 24 sample tubes, or 96-well microtiter plate format. Analysis mode: Up to 10 000 events/s, and resolution down to 0.5μm.

BD Accuri C6 Plus, +Oncyt module. Lasers: blue (488 nm), red (640 nm) Output: FSC, SSC and 4 filters Sample port: sip can be applied for any size (Eppendorf tubes to bioreactors). Equipped with an Oncyt module (http://oncyt.com/new/products-services/discrete-on-line-flow-cytometry/) for automated sampling and staining.

Digital and physical collections

All data is generated and transferred locally. The data service consists of in-house primary storage, and each registered user gets storage space on MIFLOCYT computers. The data is either stored directly to the storage system during acquisition or transferred post acquisition using custom routines. For projects with very high storage needs, MIFLOCYT reserves the right to discuss optimal storage procedures with the PI. It should be noted that MIFLOCYT does not take formal responsibility for the data stored on MIFLOCYT systems.

Services provided

MIFLOCYT is a flow cytometry platform hosted and administered by the Division of Applied Microbiology (LTH).

It is the purpose of MIFLOCYT to provide access to state-of-the art flow cytometry equipment that will allow for conducting advanced microbiological experiments, including cell population analysis and cell sorting of both pure cultures, and mixed-species communities.

The infrastructure is open to use for both LU internal and external researchers.

However, MIFLOCYT is not a core facility and we do not provide assistance with running the instruments, designing experiments or analyzing data. If this type of assistance is needed, we recommend establishing a collaboration with a researcher who is already connected to MIFLOCYT.

The researcher interested in using the instruments needs to apply for approval from the MIFLOCYT steering group to run the project within the infrastructure.

As MIFLOCYT is not a core facility, it does not provide individual services to researchers that receive approval for their project. Researchers already using MIFLOCYT can welcome new users to the platform, and can provide introductions to the booking system, the coffee room, and standard operating protocols (SOPs) required for startup, operation, cleaning and shutdown of the instrument.

Management of the infrastructure

The infrastructure is managed by the MIFLOCYT steering group consisting of experienced flow cytometry users and is appointed by the Division of Applied Microbiology to organize the use and maintain the platform.

Terms of access:

All projects performed at MIFLOCYT are evaluated before acceptance with respect to the possibility that the intended experiments can be performed successfully, with the available equipment at MIFLOCYT. This evaluation is performed by the MIFLOCYT Steering group. For this reason, emphasis in the project description should be to show that the intended experiments are possible to perform with MIFLOCYT instruments.

Future users need to fill in an application form that is available upon request. Please send an email to miflocyt@tmb.lth.se for more information. After the project has been approved, the Steering group willprovide access to the booking system.

MIFLOCYT is runon a non-profit basis. User fees are between 150-1000sek (internal), or and 250-1700sek/h (external).
