The research on which this thesis is to be based has a main focus on PCR and its underlying mechanisms. This will be reached through ways of studying the interactions and thermodynamics of the reaction by taking out and focusing on the DNA extension step using calorimetry. Further depth will be reached by studying the effect on various PCR inhibitors and the mechanisms of different inhibitors on several platforms including digital PCR in collaboration with NIST. Further, departmental studies on site at the Swedish National Forensic Centre (NFC) in Linköping will give a practical approach and training in “real-life” use of PCR and a focus will be on applying knowledge learned in solutions for the analytical chain at the biology department.
