Research

Development of immunotherapies targeting leukemia stem cells. Acute myeloid leukemia (AML) is a fatal disorder and current therapies are ineffective at defeating the disease. In most patients, the cellular origin of AML is thought to be a normal hematopoietic stem cell that has acquired genetic lesions that give rise to a pre-leukemic clone. Through a series of additional genetic lesions, the pre-leukemic clone forms cells with leukemia-initiating capacity, also termed leukemia stem cells, which propagate into frank AML. Because the leukemia stem cells are resistant to chemotherapeutic treatment, and thereby contribute to disease relapse, it is crucial to eradicate the leukemia stem cells. To developing new therapeutic approaches targeting the disease, we pioneer screens designed to identify new therapeutic targets on leukemia stem cells. We are working on the following projects:

  • In vivo pooled CRISPR/Cas9-screens using leukemia stem cells.
  • Cytokine screens using barcoded leukemia cells allowing for in vivo read-out of leukemia stem cell activity. • Small molecule screens using barcoded leukemia cells allowing for in vivo read-out of leukemia stem cell activity.
  • Proof of concept for therapeutic effect in AML mouse models using newly developed therapeutic approaches.

Successful completion of these projects will identify new therapeutic targets in AML and expand our knowledge regarding AML stem cells, findings that might be translated into new therapeutic opportunities in AML.
