Hypothesis: We propose that simple carbohydrate derivatives can be used as anti-tumor therapy, by acting as inhibitors of the glycosaminoglycan biosynthesis.
Significance: Tumor cells are highly dependent on glycosaminoglycans for proliferation, for angiogenesis and for metastasis. Thus, if we inhibit the formation of these carbohydrate structures, we will selectively target tumor cells over normal cells.
Aims: We will synthesize molecular probes to investigate the binding pocket of one of the most important enzymes in the glycosaminoglycan biosynthesis, i.e. 4GalT7. The data generated from these probes will be used for design of inhibitors that will be tested in tumor cells.
Feasibility: We have developed assays for important enzymes and built a library of carbohydrate structures. We have a very strong background in synthetic carbohydrate chemistry and in biological investigations.
