Tapasin as a prognostic and diagnostic tool in glioblastoma multiforme

HLA-I deficiency is frequent in many tumors, including glioblastoma multiforme
(GBM) and melanomas. In this project we (I) investigate the
mechanisms the effect of tapasin on different HLA-I allomorphs present in GBM patients, (II) investigate the HLA-I tapasin-dependence as a tool for prognostics and for the use in stratification for immune therapy in GBM, and possibly also in melanoma (III) study and develop a tapasin-fragment for use as an immuno-modulatory tool in vitro and in vivo against tumors with dysregulation in the cell surface expression of HLA-I (e.g., GBM and again possibly also melanomas and other types of tumors). The base for this project stems from the continuous improvement in the understanding of how tapasin function and is described in the general introduction and background. The project is at present moving forward mainly thanks to the work done by Maxim Morin. We have at the moment very promising data from biochemical and cellular models but before going into in vivo models we want to get a better understanding of how the tapasin protein act in solution (including pH dependence, concentration dependence, interaction with HLA-I and peptide). The ongoing work include expression of different tapasin recombinants in insect cells and the biochemical and biophysical characterization of its properties using scattering methods, biochemical methods as well in cellular models.
