Our team focuses on advancing the conceptual understanding for how the formation of the adaptive immune system changes with developmental timing, and in particular changes in hematopoietic stem cell (HSC) biology and its impact on B cell development and function (Yuan et al, Science, 2012; Kristiansen et al, Immunity, 2016). Using cellular barcoding , VDJ sequencing, and single cell gene expression analysis, we interrogate the ability of stem cell states to impact on downstream fate decisions and immunological diversity. Ongoing projects include:
• Interrogating the reversibility of adult HSCs to fetal-like plasticity and regenerative potential.
• Elucidating the basis for the developmental switch in B lineage output early in life.
• Stratification of the immune system based on developmental timing to visualize the division of labor between fetal and adult hematopoietic stem and progenitor cells.
