Demanding orthopedic conditions like high energy fractures, infection, tumors, non-unions, mal-unions require additional bone at surgery. This has conventionally been done using bone grafts either taken from the patient him/herself (Autograft) or from deceased donors (Allografts). Grafting techniques incur drawbacks like donor site morbidity and inadequate volume of bone grafts in the case of autografts. Likewise, allografts pose a risk of disease transmission as well as their efficacy varies from donor to donor. Our research primarily focuses on developing biomaterials either ceramic or composites of ceramic and polymeric materials, which are biocompatible and safe for clinical use. A limitation of biomaterials is their limited osteoinductivity i.e. their ability to induce bone formation by the differentiation of osteoblasts precursors into bone forming osteoblasts. This adds another dimension to our research. By adding growth factors from the potent bone morphogenic protein (BMP) family such as BMP-2 and BMP-7, we enhance the osteoinductivity of the scaffolds. We also deliver other bone active molecules like bisphosphonates to further supplement the bone growth. Thus, our research primarily deals with 1) Developing novel bone mimicking biomaterials 2) Using these biomaterials as a platform for controlled spatio-temporal delivery of bone active molecules that can accelerate bone regeneration. In essence, we explore the principles of biomaterial sciences, drug delivery and tissue engineering. Most of the laboratory work is being performed on small animals at the moment and our laboratory has developed a couple of novel models to study bone regeneration in healthy as well as osteoporotic rats.
